Next-Generation Tuberculosis Vaccine Research and Treatment Strategies

Tuberculosis (TB) remains a major global health challenge in 2026, particularly with the rise of multi-drug-resistant (MDR-TB) strains. While the BCG vaccine has been used for over a century, its efficacy in adults is limited. Current research is focused on "Prime-Boost" strategies, where the BCG vaccine is followed by a booster of a recombinant protein or viral-vector vaccine.

The leading candidate in 2026, M72/AS01E, has shown significant promise in preventing active TB in individuals already infected with the bacteria. Treatment strategies for active TB have also evolved, with "Short-Course" regimens (4 months instead of 6) becoming the standard. These new protocols utilize potent combinations of bactericidal agents that can penetrate the thick waxy wall of the Mycobacterium tuberculosis, improving compliance and reducing the emergence of resistance.
